Description

eDNA is an exciting new development for the detection of environmental DNA to look for the presence of specific organisms in a water body. It works by taking water samples from the site and sending them to a specialised laboratory which extracts and amplifies the DNA of the target species. It is a very specialised analysis with a high level of accuracy, and potentially has a wide range of applications. What it can do is find minute traces of the target organisms with a high degree of accuracy.

Fish Treatment Ltd, in collaboration with Mainstream Fisheries Ltd, has been involved in the development of unique eDNA testing both in the Coarse Fish Industry and in Ornamental fish, and have acquired exclusive access to a state-of-the-art specialist laboratory.

Specific disclaimer and general information

  1. In general, the larger the sample volume the more accurate the eDNA detection levels.
  2. Our recommendation is to use our laboratory results as an indication to be confirmed with physical examination of infected fish.
  3. Our eDNA laboratory results will have a confidence level of at least 95%.
  4. There are a number of factors which may affect the results, including biological variations, season, water temperature etc.

DNA Swabbing

  1. Put on gloves
  2. Hold the swab on the plastic end. Do not touch the swab end.
  3. Rub the swab end on to the surface you wish to take the same from while rotating the swab between your finger and thumb at the same time.
  4. Unscrew the blue cap on the tube.
  5. Place the swab, swab end first into the preservation tube.
  6. Snap the plastic stem by moving the stem to one side.
  7. Screw the blue cap back on the preservation tube.
  8. Use the SAE to return the preservation tube with your sample and test request.

Collecting the water samples

Overview

The samples for testing are collected by passing water which the fish inhabit through a filter. The water is discarded after passing through the filter – it is the contents of the filters that will be tested when they are returned to us. Below is a list of items you will require to collect the samples in the filters and step by step instructions describing how to collect the samples. Please follow these instructions carefully so we receive the correct quantity of material to test and thus ensuring the collected samples are in good condition. One sample kit tests one body of water, and the gloves and syringes should be discarded after use.

Water sampling kit Per 500ml of water

  1. 1 x Padded jiffy bag with correct postage for return.
  2. 1 x Cooling gel pack
  3. 4 x Sterile Blue gloves
  4. 1 x Clear Grip Seal bag
  5. 1 x 50ml Luer-lock syringe
  6. 1 x 0.22µm Sterivex filter
  7. 2 x Sterivex filter red caps (for Inlet and Outlet)
  8. 1 x Barcoded labels
  9. 1 x Submission form

Sampling protocol

  1. Put cooling gel pack in the freezer for at least 2 hours before you begin sample collection.
  2. With clean dry hands put on the sterile blue gloves before you start.
  3. Remove the packaging from a syringe and a filter.
  4. Fill the syringe with water from the body of water which is being tested to the 50ml mark.
  5. Screw the syringe to the inlet of the filter (plain long end). Pass the water through the filter slowly by carefully pushing the syringe plunger. Remove the filter from the syringe.
  6. Repeat steps 4 and 5, ten (10) times, this ensures we have enough material to test. Please note towards the end it will get more difficult to pass the water from the syringe through the filter.
  7. Once you have done this, disconnect the syringe from the filter and fill with air by pulling up the plunger, re-connect to the filter inlet and push air through the filter. Repeat this step until no water is visible in the filter. Remove and discard the syringe.
  8. Cap the filter at both ends with the red caps provided.
  9. Stick the barcoded label on the side of the filter.
  10. Insert the filter into the clear grip seal bag.
  11. Repeat steps 3 to 10 for each of the 2 filters.
  12. The filters should be stored at a cool temperature until shipping for processing, ideally in a refrigerator at 4⁰C.
  13. Write the barcode, number of samples, your details and the sampling date on the submission form.
  14. For shipping, insert the Grip Seal bag containing the filters, submission form and frozen gel pack into the padded envelope provided and post back to eDNA Mainstream within 48 hours of taking the sample.

Testing Terms & Conditions

Reference to liability in this clause include every kind of liability arising under or in connection with the Agreement including liability in contract, tort (including negligence), misrepresentation, restitution or otherwise.

The Customer understands that there are a number of factors that may affect the Analysis Result, including biological variations and mutations, as well as the choice of processing circumstances upon receipt of the Analysis Results, such as season, water temperature, etc., and the Customer is responsible for actions, omissions or measures executed by the Customer or a third party.

The Supplier is not responsible for how Analysis Results are used by the Customer or any third party to whom the Customer provides the Analysis Results, and the Customer is always encouraged to confirm or refute any suspicions with at least two methods, and with a sufficient statistical selection.

The Supplier is not responsible for any errors, delays or deficiencies due to, in whole or in part, circumstances arising during registration of samples, transport, contamination of samples or non-compliance with the instructions provided by the Supplier.

The Supplier is not responsible for indirect, special, incidental or unpredictable losses, or consequential losses of any kind, including losses due to service disruptions, continued/inappropriate operations or lost profits, loss of sales or business and loss of agreements or contracts.

This clause shall survive termination or expiry of the Agreement.
